چکیده
Abstract Alveolar macrophages (AMs) are less able to respond IL-4 in vivo than from the peritoneal cavity, due a still-unknown factor of lung environment. The aim this study is investigate whether surfactant lipids, which continuously endocytosed by AMs, could influence IL-4-mediated alternative activation and proliferation AMs. To that end, AMs were preincubated with lipids stimulated presence or absence protein SP-A, an amplifier actions. We found alveolar reduced IL-4- IL-4+SP-A-dependent arginase activity, expression genes associated activation, Mechanistically, decreased IL-4+SP-A-induced PI3K-Akt-mTORC1 signaling axis, but not IL-4-dependent STAT6 axis. Lipid-dependent inhibition Akt/mTORC1 axis consistent IL-4+SP-A-driven glycolysis mitochondrial respiration as well ATP citrate lyase histone acetylation IL-4. conclude inhibit PI3K-dependent pathways suggest decrease chronic diseases might augment fibrotic responses. Insulin inhibits LPS-induced signaling pathways in alveolar macrophages.
The systemic inflammatory response syndrome (SIRS) is triggered by lipopolysaccharide (LPS) from Gram-negative bacteria. Insulin was shown to have a protective role in SIRS related to sepsis. Lungs are particularly affected in this condition and provide a second wave of mediators/cytokines which amplifies SIRS. The aim of the present study was to investigate the effect of insulin on the signali...
متن کاملIL-4 Can Inhibit IL-17 Production in Collagen Induced Arthritis
Background: IL-4 is a cytokine that induces differentiation of naive helper T cells into Th2 cells. Once activated by IL-4, Th2 cells subsequently produce additional IL-4. Objective: To examine the effect of IL-4 on IL-17 production and its effect in Collagen- Induced Arthritis (CIA) mice. Method: In this study, a chicken collagen-II-induced experimental arthritis (CIA) model was used in DBA/1 ...
متن کاملIn vivo and in vitro uptake of surfactant lipids by alveolar type II cells and macrophages.
The uptake of fluorescent-labeled liposomes (with a surfactant-like composition) by alveolar macrophages and alveolar type II cells was studied using flow cytometry, in vivo by instillation of the labeled liposomes in the trachea of ventilated rats followed by isolation of the alveolar cells and determination of the cell-associated fluorescence, and in vitro by incubation of isolated alveolar c...
متن کاملRAGE signaling by alveolar macrophages influences tobacco smoke-induced inflammation.
Receptors for advanced glycation end-products (RAGE) are multiligand cell surface receptors of the immunoglobin family expressed by epithelium and macrophages, and expression increases following exposure to cigarette smoke extract (CSE). The present study sought to characterize the proinflammatory contributions of RAGE expressed by alveolar macrophages (AMs) following CSE exposure. Acute exposu...
متن کاملRole of clathrin-mediated endocytosis of surfactant protein A by alveolar macrophages in intracellular signaling.
We recently provided evidence that anti-inflammatory macrophage activation, i.e., the inhibition of constitutive and signal-induced NF-kappaB activity by the pulmonary collectin surfactant protein (SP)-A, critically involves a promoted stabilization of IkappaB-alpha, the predominant inhibitor of NF-kappaB, via posttranscriptional mechanisms comprising the activation of atypical (a)PKCzeta. SP-A...
